چکیده
Neochamaejasmin A is an individual compound isolated from Stellera chamaejasme and exerts antitumor effects on several types of human cancer cells. However, the effect neochamaejasmin osteosarcoma cells has not yet been investigated. The main aim was to detect in vitro MG-63 were treated with different concentrations (20, 40, 80 μg/ml). Cell viability detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. apoptosis through cell morphology observation Annexin V/propidium iodide staining. Mitochondrial membrane potential measured rhodamine 123 expressions apoptosis-related proteins phosphatidylinositol 3 kinase/protein kinase B signaling analyzed western blotting. results showed that significantly inhibited proliferation induced MG- 63 triggered disorder mitochondrial potential, increased expression Bcl-2-associated X protein cleaved-caspase 3, decreased B-cell lymphoma 2, inactivation B/glycogen synthase kinase-3-beta pathway. These new findings may indicate be chosen as a candidate for chemotherapy.

Inhibition of autophagy attenuated curcumol-induced apoptosis in MG-63 human osteosarcoma cells via Janus kinase signaling pathway
The present study aimed to investigate whether autophagy was triggered by curcumol and to explore the association between autophagy and apoptosis of MG-63 cells and the underlying mechanism. MG-63 cells were cultured in vitro. An MTT assay was performed to evaluate the proliferation inhibition of the MG-63 osteosarcoma cell line by curcumol. Fluorescein isothiocyanate-Annexin V/propidium iodide...
متن کاملberberine induced apoptosis of human osteosarcoma cells by inhibiting phosphoinositide 3 kinase/protein kinase b (pi3k/akt) signal pathway activation
background: osteosarcoma is a malignant tumor with high mortality but effective therapy has not yet been developed. berberine, an isoquinoline alkaloid component in several chinese herbs including huanglian, has been shown to induce growth inhibition and the apoptosis of certain cancer cells. the aim of this study was to determine the role of berberine on human osteosarcoma cell lines u2os and ...
متن کاملInhibition of focal adhesion kinase induces apoptosis in bladder cancer cells via Src and the phosphatidylinositol 3-kinase/Akt pathway
Focal adhesion kinase (FAK) is a 125-kDa, cytosolic, non-receptor, protein tyrosine kinase localized at focal adhesions that can be activated by multiple inputs and in different manners. FAK is implicated in signaling pathways regulating cell movement, invasion, survival, gene expression and cancer stem cell self-renewal. The aim of the present study was to investigate whether FAK plays a role ...
متن کاملTrastuzumab increases pulmonary vein arrhythmogenesis through modulating pulmonary vein electrical and conduction properties via phosphatidylinositol 3-kinase signaling
Objective(s): Drug-induced atrial fibrillation (AF) is considered an adverse effect of chemotherapeutic drugs. AF is a crucial risk factor for stroke, heart failure, myocardial infarction, and mortality. Pulmonary veins (PVs) are considered triggers inducing AF, and the sinoatrial node (SAN) may modulate PV activity and participate in AF genesis. AF was associated with...
متن کاملPhosphatidylinositol 3-kinase (PI3K) signaling via glycogen synthase kinase-3 (Gsk-3) regulates DNA methylation of imprinted loci.
Glycogen synthase kinase-3 (Gsk-3) isoforms, Gsk-3α and Gsk-3β, are constitutively active, largely inhibitory kinases involved in signal transduction. Underscoring their biological significance, altered Gsk-3 activity has been implicated in diabetes, Alzheimer disease, schizophrenia, and bipolar disorder. Here, we demonstrate that deletion of both Gsk-3α and Gsk-3β in mouse embryonic stem cells...
